    Title: 以逢機增殖多型性DNA (RAPD)進行火鶴花栽培種鑑定與遺傳歧異性之研究

    Abstract: 本研究以農業試驗所「火鶴花育種計畫」中收集的15個栽培種為材料,探討利用逢機增殖多型性DNA(randomly amplified polymorphic DNA;RAPD)分子技術作為品種鑑定及遺傳歧異性分析。試驗結果顯示,自8條含有10個核甘酸的逢機引子經聚合酵素連鎖反應(polymerase chain reaction ; PCR)產物中,有24個特定的DNA多型性標誌片段,可用以鑑別出14個栽培種。進一步以此8個多型性表現高的引子,將其PCR反應產物160個RAPD片段,合併進行UPGMA(unweighted pair-group mean arithmetic)群集分析,結果獲得三個主要群組。第一群包含8個栽培種,其佛燄苞顏色(花色)主要為紅色系。第二群為6個栽培種組成,主要為源自夏威夷育成之品種。而第一、二兩群14個栽培種均屬Anthurium andraeanum。第三群則僅有一個獨立的栽培種“Southern Blush”,為A. andraeanum X A. amnicola之種間雜交種。試驗結果顯示,利用RAPD兼具有鑑別火鶴花栽培種之能力,亦可用以估測品種間基因型之遺傳相似性,此對於未來藉以選拔遺傳歧異高的雜交親本及新雜交組合的獲得,將有所助益。整體而言,RAPD分析技術對於加速火鶴花育種效率,應具有實用價值。
